【Pharmacological Effects】Pharmacodynamics: Penicillin is a bactericidal antibiotic with strong antibacterial activity. Its antibacterial mechanism mainly involves inhibiting the synthesis of the cell wall glycopeptide in bacteria. During the growth phase, sensitive bacteria divide vigorously, and the cell wall is in the biosynthetic stage. Under the action of penicillin, the synthesis of glycopeptide is blocked, preventing the formation of the cell wall. Under the pressure of osmotic pressure, the cell membrane ruptures and the bacteria die.

Penicillin is a narrow-spectrum antibiotic, mainly exerting effects on various Gram-positive bacteria and a few Gram-negative cocci. The main sensitive bacteria include Staphylococcus, Streptococcus, Brucella suis, Corynebacterium, Clostridium tetani, Actinomyces, Pyogenic bacteria, and Spirochetes, etc. It is not sensitive to Mycobacterium, Mycoplasma, Chlamydia, Rickettsia, Nocardia, fungi and viruses.

Pharmacokinetics: After intramuscular injection of procaine penicillin, penicillin is released through local hydrolysis and then slowly absorbed. The peak time is long, the blood concentration is low, but the effect lasts longer than that of penicillin. It is limited for use in pathogenic bacteria that are highly sensitive to penicillin and should not be used for treating severe infections. When procaine penicillin is mixed with penicillin sodium (potassium) to prepare an injection, it can increase the blood drug concentration in a short period of time, balancing long-lasting and rapid effects. Large doses of procaine penicillin can cause procaine poisoning.

【Drug Interactions】 (1) When penicillin is used in combination with aminoglycoside antibiotics, it can increase the concentration of the latter in the bacterial cells, thus resulting in a synergistic effect.

(2) Rapid-acting antibacterial agents such as macrolides, tetracyclines and aminoglycosides can interfere with the bactericidal activity of penicillin and should not be used in combination.

(3) Heavy metal ions (such as copper, zinc, and mercury), alcohols, acids, iodine, oxidants, reducers, hydroxyl compounds, acidic glucose injection solution or tetracycline hydrochloride injection solution, etc., can destroy the activity of penicillin and are considered as contraindications for compatibility.

(4) Some drug solutions (such as chlorpromazine hydrochloride, lincomycin hydrochloride, tartrate norepinephrine, tetracycline hydrochloride, tetracycline base hydrochloride, B vitamins and vitamin C) should not be mixed together, otherwise turbidity, flocculent substances or precipitation may occur.

【Indications】It is mainly used for chronic infections caused by penicillin-sensitive bacteria, such as retained placentae in cows, mastitis, complex fractures, etc., and also for infections caused by actinomycetes and leptospira, etc.

【Usage and Dosage】Intramuscular injection: The dosage is 10,000 - 20,000 units per 1 kg of body weight for horses and cattle; 20,000 - 30,000 units for sheep, pigs, foals, and calves; 30,000 - 40,000 units for dogs and cats. Administer once daily for 2 - 3 days.

Before use, add an appropriate amount of sterilized injection water to form a suspension.

【Adverse Reactions】(1) The main type of reaction is an allergic reaction, which can occur in most domestic animals, although the incidence is relatively low. Local reactions manifest as swelling and pain at the injection site, while systemic reactions include hives and rashes. In severe cases, it can lead to shock or death.

(2) For some animals, it can induce secondary infections in the gastrointestinal tract.

【Notes】 (1) This product is only used for treating chronic infections caused by highly sensitive bacteria.

(2) It is slightly soluble in water. It will rapidly lose its efficacy upon contact with acids, alkalis, oxidants, etc. Therefore, the injection solution should be prepared immediately before use.

(3) Pay attention to the interactions and contraindications with other drugs to avoid affecting the efficacy of the medication.
