【Pharmacological Effects】Pharmacodynamics: Triclabendazole belongs to benzimidazole drugs and is specifically used for the treatment of fasciolopsis buski. It has a significant killing effect on various age groups of liver fasciolopsis buski and is a relatively ideal drug for killing liver fasciolopsis buski. After absorption, it interferes with the microtubule structure and function of the parasite, inhibiting the release of proteolytic enzymes in the parasite. The effect of triclabendazole on the parasite varies with the concentration. For adult worms, they can survive for 24 hours in a low concentration (1-3 pg/ml) of the drug, while in a higher concentration (10-25 μg/ml), their activity significantly weakens after 24 hours; in a high concentration (25-50 μg/ml), they are completely inhibited after 24 hours. However, it is more sensitive to larval worms, and at a concentration of 10 μg/ml, their activity is completely inhibited after 24 hours. Pharmacokinetics: The bioavailability of triclabendazole is high. When goats and sheep are orally administered a dose of 10 mg/kg body weight, the peak blood concentration reaches 15 ug/ml within 24-36 hours. The peak blood concentrations of triclabendazole and its metabolites are 5-20 times higher than those of other benzimidazole anthelmintics. The elimination half-life is approximately 22 hours. Triclabendazole is mostly oxidized into sulfoxides and sulfones in the bodies of sheep and rats, and these derivatives bind to albumin and remain in the plasma for more than 7 days. High plasma concentrations and the binding of drugs to plasma albumin seem to be related to the prolongation of the action time of anthelmintic drugs against fasciolopsis buski. After 10 days of administration in sheep, approximately 95% of the drug is excreted in feces, 2% through urine, and less than 1% through milk.
